Anti-Jo-1 antibody is a type of autoantibody that targets histidyl tRNA synthetase, an enzyme involved in protein synthesis. It is associated with a group of autoimmune diseases collectively known as idiopathic inflammatory myopathies (IIMs), with the most common being polymyositis and dermatomyositis.

Detection of anti-Jo-1 antibodies is significant in diagnosing these autoimmune conditions, particularly when patients present with muscle weakness, skin rash, and other symptoms suggestive of myositis. Anti-Jo-1 antibody testing is typically performed as part of a panel of tests to help confirm the diagnosis of these inflammatory myopathies. Treatment usually involves immunosuppressive medications to manage symptoms and suppress the autoimmune response.
